Two types of microscopes used in immunofluorescent techniques are the
t_______d l___t microscope and the i____t light or e__-i_______n microscope.
One major difference is the d____n from which the light or energy strikes the
specimen. In the transmitted light microscope, light strikes the specimen from
(above) (below) through a c______r. In the incident light microscope, light strikes
the specimen from (above) (below), passing through the o___ve; this eliminates
the need for a c___r and also eliminates the problems of c____g a condenser.
The incident microscope has more b____ness, clearer i___ges, and greater
f____e since illumination and observation of the specimen are made from the
same d_____n.

The component parts of the two types of microscopes include the l___t system
and the f___r system.
The light system includes the light r_____r, the light s____e, and the collecting
l__ses. The filter system includes the h__t-absorbing filter, the e___r filter, the
dic___c mirror, and the b___r filter.
The light reflector is a concave m____r located behind the l___t s___e which
redirects l__t energy (light) back into system. The light source is a mercury v___r
lamp. The collecting lenses c______rate light from the light source into a single
b___m.
The heat-absorbing filter removes excess h__t from the exciting light that may
damage the system. The exciter filter (primary filter) transmits only the e____ve
light and supp_____s all other energy from light source emission which are not
required for s____n fluorescence. The dichroic mirror, which is part of the i____t
microscope only, allows passage of light of selected wave l____s in one direction
through the mirror but not in the o____e direction. The barrier filter transmits only
the e___ted fluorescent light from the specimen and suppresses all other e___y.
